SIMPLE STAINING
INTRODUCTION:
The coloration of bacteria by applying a single staining solution to a fixed smear is a “simple staining”. It is called as “monochrome staining” because only one dye is used. Basic dyes like methylene blue, crystal violet, safranine, malachite green are used to determine the shape, size and arrangement of bacteria.
PROCEDURE:
➤ Prepare the smear and heats fix it.
➤Flood the smear with methylene blue stain and allow it to react for 1 minute.
➤Wash the slide with distilled water, blot and air dry.
➤ Examine the slide under microscope.
RESULT:
➥ Organisms are stained blue in color.
